The film opens with the French national soccer team’s victory over the Chinese national soccer team, the murder of French coach Yves Gluant (Jason Statham), and the disappearance of his Pink Panther diamond ring. Chief Inspector Charles Dreyfus (Kevin Kline), eager to win the Légion d'honneur (translated as the "Medal of Honor" in the film), appoints the "most incompetent policeman" in France, Inspector Jacques Clouseau (Steve Martin), to the case as a diversion. Dreyfus, meanwhile, assembles a team of investigators to tackle the mystery in secret, planning to take over the case and win the Légion d'honneur. Dreyfus also appoints Gendarme Gilbert Ponton (Jean Reno) to report on Clouseau's whereabouts at all times.

La Panthère Rose, l'extraordinaire diamant, vient d'être volé, et son propriétaire, le sélectionneur de l'équipe nationale a été assassiné. Une magnifique pop star (Beyoncé) ? Un footballeur ? Un dignitaire chinois ? Les pistes sont nombreuses. Le gouvernement français a plus que jamais besoin d'un détective hors-pair pour démasquer le meurtrier et retrouver le célèbre joyau. Le fourbe Dreyfuss (Kevin Kline), décidé à s'attribuer tous les honneurs de la résolution finale, confie l'enquête à son calamiteux subalterne, l'Inspecteur Clouseau (Steve Martin). Multipliant les catastrophes, Clouseau, aidé du taciturne Ponton (Jean Réno), ne se rapproche pas moins de l'odieux malfaiteur.
