Eastwood (marketed as the "Man with No Name") and Van Cleef (as Colonel Douglas Mortimer and marketed as "The Man in Black") portray two bounty hunters in pursuit of "El Indio" (Gian Maria Volontè), one of the most wanted fugitives in the western territories, and his gang (one of whom is played by Kinski). Indio is a ruthless, intelligent man addicted to smoking cannabis. He has a musical pocketwatch that he plays whenever he wishes to kill someone. "When the chimes finish, begin," he says, and sure enough when the music stops he shoots his victim. Through flashbacks we learn that Indio's pocketwatch (and perhaps cannabis addiction) originates from a young woman who killed herself while being raped by Indio, after Indio finds her with her lover and kills him out of jealousy.
Deux chasseurs de primes (Eastwood et Van Cleef) traquent un pilleur de banques extrêmement dangereux (Volonte). Après "Pour une poignée de dollars", ce deuxième épisode de la trilogie qui s'achèvera avec "Le Bon, la Brute et le Truand" n'est pas une suite mais met en scène le même genre de personnages animés par un seul appât, celui du gain. L'univers reste le même : les grands espaces sont filmés à la loupe (à Almeria, en Espagne), les dialogues n'excèdent jamais l'essentiel, l'humour joue la noirceur, la sensualité est morbide et l'indispensable Ennio Morricone s'applique à ponctuer toute l'histoire de sa petite mélodie envoûtante. Sergio Leone, pourtant, implique davantage ses personnages : la relation entre Eastwood et Van Cleef est loin d'être vaine, les codes d'honneur sont omniprésents et l'amitié n'est pas loin. En plus de celle de Lee Van Cleef, véritable gueule du cinéma américain, on signalera la présence de l'acteur allemand Klaus Kinski qui affichait d'emblée un génie insolent et un visage inoubliable.
